No, you don't need to drain any fluid.
You may get a little differential oil leaking out the wheel end of the axle housing, depending on what angle you have the axle supported at. No big deal. The bigger deal is to make sure you don't drag the axle along the bottom of the housing upon reinstallation. There's a bunch of crud that can get pushed into the axle seal and differential. Not good.
